Please go to the Allen Cell Explorer homepage and familiarize yourself with the site, the kinds of data it contains , and some of the interfaces. The Allen Cell Collection contains over 50 human stem cell lines that have been genetically modified to express a fluorescently tagged version of a protein found in key cellular structures and sub-structures . Each of these cell lines has been extensively imaged during cell division using confocal microscopy , and Al algorithms have been used to form consensus images of how different labeled cell structures behave at different stages . We will not use the full resources that it makes available but we will use some of them to better understand cell biology . In this assignment , we will use an interface called the Integrated Mitotic Stem Cell https://imsc.allencell.org/?page=1& ga2,145464381.885795841.1678286759- 1907298573.1678138161 . Scroll through the Integrated Mitotic Stem Cell interface . For me, it scrolls forward easily if I put my cursor over where it says scroll but if you want to go back it helps to click on the progress bar at the top When you get to the end you will be in the Explore in 3D section . You will use the 3D Cell Viewer here to complete your assignment which should be handed in as a single word document containing the following : Part 1 (10 points ) : Use the 3D Cell Viewe to label only the DNA and microtubules (TUBA1B ) at all stages of mitosis shown . Select the option “Raw” for image processing , and feel free to zoom in until the cell almost fills the screen . Take a screen capture of the cell in each of the cell stages listed (Interphase , Prophase Prometaphase , Metaphase , and Anaphase ) and paste them into a word document and label them .
Part 2 (10 points): Go to the drop-down menu labeled ” Pathtrace renderings for structures that:” which can be found just to the left of the button that says “Raw.” In your homework file, provide a labeled image of a cell in Interphase, Metaphase, and Anaphase for each of the three categories : Redistribution & Reorganization Disassembly & Recompartmentalization Maintained This means that for part 2 you should have nine images in total. Note what tagged genes are being shown in each category .
