This semester, you and your colleagues taking Genetic Techniques used DNA barcoding to identify mosquitoes (or other organisms) and PCR screening to determine if those organisms harbored the Wolbachia endosymbiont. Think of yourselves as a research team all working together to put together datasets that can be used to address a single research objective. You will analyze and interpret the aggregate data generated from all members of the team, not just your personal results. The two datasets include the COI DNA sequences and a table of positive or negative screening results for Wolbachia.
Analyze this data using the relevant analytical techniques that you have learned in this course. These include measures of genetic distance (within and between species) and a phylogenetic tree. The raw presence/absence table for Wolbachia (the overall class results which I provide to you as a spreadsheet) must be summarized using an appropriate figure or table (do not copy/paste the spreadsheet into your report!). If you feel there are other analytical techniques that can be used to address your research objective, feel free to use them–but they must be relevant.
3-4 pages of text plus additional pages containing figures and/or tables.
Do not include a cover sheet.
Double spaced, 1-inch margins, Times New Roman font, 12 pt, single column of text.
Figures/tables must be of your own creation (not copied and pasted) and must only contain results (but do not include images of your gels). Do NOT include in your report the raw DNA sequences or spreadsheet provided to you! Tables and figures show summaries of the raw data, not the raw data itself.
Use the research papers that you have read (and those provided as examples) as a guide to scientific writing. You need to study these papers carefully and mimic them. The closer your paper is to the scientific style of writing, the better your grade will be.
Start with a title followed by your name and then the following three sections (use headings):
Introduction: Prepare the reader so that they can understand the context of your research objective (which must be clearly stated in the last sentence of the Introduction). Approx one page.
Methods: Use complete sentences to state what methods you used and the specific purpose of each (in the order you performed them). Do not include protocols, but do include the final concentration of PCR reagents (this is the concentration of reagents within the reaction after they are all mixed together, not the concentration of each stock reagent), details of the PCR thermal profile (the information you need for this is provided), and what primers were used. That information can be found here (Links to an external site.). Be sure to state what primers were used, their DNA sequence, and the predicted size of the PCR products (drawing from resources provided). Since this is a techniques course, I’ll expect that you do an amazing job on this section. The majority of your grade for the paper will be based on how well you write this section. Approx two pages.
Results & Discussion: State the results (referencing any tables and/or figures of your own creation) and interpret them in light of your research objective. Approx one page.
Literature Cited: You do not have to cite the literature as extensively as the example papers, but do include at least two citations to appropriate scientific literature in the Introduction and/or Discussion.
Figures/Tables: Place these on a separate page (or pages) at the very end. Each figure and table must have a caption so that the figure/table can be understood without having to read the paper. Figure captions always go below the figure. Table captions always go above the table. See the example papers for examples of how to write appropriate captions.
Note: the scientific name of organisms must must be formatted properly. Species names are binomials (consist of two parts, the generic epithet and the specific epithet). Both names must be italicized, the genus capitalized, and the species lowercase (e.g. Homo sapiens). This is a universal format, and others will view you as ignorant and incompetent if you do not apply it in all future situations.
