X-ray microscopy reminds me of another lab technique called x-ray crystallography. In x-ray crystallography a 3-D structure is formed instead of a 3-D model, however, both of them are exposed by an x-ray beam. With X-ray crystallography the sample needed to be purified, crystalline, and highly concentrated. The x-rays would be diffracted allowing for information about crystal packing symmetry and size of the repeating unit of the crystal. The spot intensity is used to find structure factors, ultimately a thermodynamically favored conformation is formed. What more can you tell me about x-ray crystallography?
